Antioxidant Activity and Neuroprotective Effect of Root Bark of Morus alba L. Extract against Hydrogen Peroxide-Induced Cytotoxicity in PC12 Cells

Abstract
In this study, the antioxidant activity and neuroprotective effect of root bark of Morus alba L. on H2O2-induced differentiated PC12 cells were investigated. In order that, total phenolic and flavonoid contents of Morus alba L. extract in different ethanol concentrations (0, 30, 50, 70, and 100%) were measured. Antioxidant activities has been evaluated by oxygen radical absorbance capacity assay, 1,1-diphenyl-2-picrylhydrazyl radical scavenging assay, 2,2¡¯-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging assay, ferric reducing antioxidant power assay, and Fe2+ chelate assay. The results suggested that 70% ethanol extract of root bark of Morus alba L. had high phenolic and flavonoid contents (35.0¡¾1.5 mg/g, 7.9¡¾0.2 mg/g) and antioxidant activities. The 70% ethanol extract of root bark of Morus alba L., which showed the most antioxidative activities, was treated on H2O2-induced differentiated PC12 cells in order to evaluate its cell protective effect by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay and cell membrane protective effect by lactate dehydrogenase (LDH) assay. The results showed that cell viability was increased when treated with 25¡­200 ¥ìg/mL for 70% ethanol extract of root bark of Morus alba L., and LDH amount was decreased in a dose dependant manner. The results of this study demonstrate that Morus alba L. has potent antioxidant and neuroprotective effects against H2O2-induced oxidative stress.
